10X Phosphate Buffered Saline (PBS)

NOTE: This is to make stock 10X PBS. 1X PBS, as needed for DAPI, is located below.


Reagents (for 1L):

  • NaCl (Fisher, BP358-212) - 80g
  • KCl (Sigma, P9541-500G) - 2.0g
  • Na2HPO4:7H2O – 14.4 g
  • KH2PO4 (Sigma P3786-500G) - 2.4g
  • Pure Water H2O - 1L

Procedure:

  1. Add each of reagents to 800 mL of pure water.
  2. Adjust by adding enough pure water to make 1000mL of solution.
  3. The pH should be between 7.2 and 7.6;adjust if necessary.
  4. Autoclave and Store at Room Temperature

1X PBS

Reagents (for 1L):

Materials:

  •  50 mL presterilised centrifuge tube (VWR Cat. No. 89004-364)

Procedure:

  1. Take 5mL of the 10x PBS and place in a 50 mL centrifuge tube.
  2. Add 45 mL of sterile H2O.
  3. Mix thoroughly.
